چکیده

The sixth PDZ domain from GRIP1 and its complex with the octapeptide of the liprin-alpha1 C-terminus were crystallized at 294 K by the hanging-drop vapour-diffusion method. The native crystal belongs to space group P6(1)22 (or P6(5)22), with unit-cell parameters a = b = 40.3, c = 222.9 A. The complex crystal belongs to space group R32, with unit-cell parameters a = b = 117.8, c = 102.0 A. Native and peptide-complex diffraction data were collected to resolutions of 1.5 and 1.8 A, respectively, using synchrotron X-rays.

Crystallization and preliminary X-ray crystallographic studies of the PDZ domain of Shank1 from Rattus norvegicus.

Shank proteins are a new family of scaffold proteins interacting with various membrane and cytoplasmic proteins. Shank contains multiple protein-protein interaction sites, including ankyrin repeats, an SH3 domain, a PDZ domain, a long proline-rich region and an SAM domain. The PDZ domain of Shank binds to the C-terminus of guanylate kinase-associated protein (GKAP). The PDZ domain of Shank1 fro...

Crystallization and preliminary X-ray crystallographic analysis of the C-terminal domain of guanylate kinase-associated protein from Rattus norvegicus.

Guanylate kinase-associated protein (GKAP) is a scaffolding protein that plays a role in protein-protein interactions at the synaptic junction such as linking the NMDA receptor-PSD-95 complex to the Shank-Homer complex. In this study, the C-terminal helical domain of GKAP from Rattus norvegicus was purified and crystallized by the vapour-diffusion method. To improve the diffraction quality of t...

PDZ7 of glutamate receptor interacting protein binds to its target via a novel hydrophobic surface area.

Glutamate receptor interacting protein 1 (GRIP1) is a scaffold protein composed of seven PDZ (Postsynaptic synaptic density-95/Discs large/Zona occludens-1) domains. The protein plays important roles in the synaptic targeting of alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors. The interaction between GRIP1 PDZ7 and a Ras guanine nucleotide exchange factor, GRASP-1, reg...

Crystal structure of GRIP1 PDZ6-peptide complex reveals the structural basis for class II PDZ target recognition and PDZ domain-mediated multimerization.

PDZ domains bind to short segments within target proteins in a sequence-specific fashion. Glutamate receptor-interacting protein (GRIP)/ABP family proteins contain six to seven PDZ domains and interact via the sixth PDZ domain (class II) with the C termini of various proteins including liprin-alpha. In addition the PDZ456 domain mediates the formation of homo- and heteromultimers of GRIP protei...

Epidermolysis bullosa and embryonic lethality in mice lacking the multi-PDZ domain protein GRIP1.

Glutamate receptor-interacting protein 1 (GRIP1) is an adaptor protein composed of seven PDZ (postsynaptic density-95/Discs large/zona occludens-1) domains, capable of mediating diverse protein-protein interactions. GRIP1 has been implicated in the regulation of neuronal synaptic function, but its physiologic roles have not been defined in vivo. We find that elimination of murine GRIP1 results ...
